چکیده

We demonstrate that an exact analytical solution can be constructed for an intracellular directed transport model ofintegrin developed by Schmidt et al. (1994) The model attempts to mimick the experimental observation that integrin is transported intermittently, by a combination of two-dimensional diffusion and cytoskeleton-mediated convective transport, towards the cell edge In particular, the model assumes stochastic coupling and uncoupling of integrin molecules (described by first-order rate coefficients kc and ku) to a cytoskeletal element moving at a fixed velocity Vo Uncoupled integrins are assumed to undergo isotropic twodimensional diffusion with a diffusion coefficient Do We demonstrate for this model that, in the asymptotic limit of transport over large distances and long times, transport is described by parallel diffusion and convection processes with effective diffusivity D = DOKD/(1+ KD) and effective velocity V = Vo/(1+KD), where KD " ku/kc is an equilibrium constant for decoupling. At shorter times~ the mean-squared displacement cannot be described by superposing diffusion and convection; rather complicated transport arises from dynamical correlations associated with the coupled reaction, diffusion~ and convection

Integrin-cytoskeletal interactions in migrating fibroblasts are dynamic, asymmetric, and regulated

We have used laser optical trapping and nanometer-level motion analysis to investigate the cytoskeletal associations and surface dynamics of beta 1 integrin, a cell-substrate adhesion molecule, on the dorsal surfaces of migrating fibroblast cells. A single-beam optical gradient trap (laser tweezers) was used to restrain polystyrene beads conjugated with anti-beta 1 integrin mAbs and place them ...

Integrin dynamics on the tail region of migrating fibroblasts.

Cell migration is a complex process that can be considered as a repeated cycle of lamellipod extension and attachment, cytoskeletal contraction, and tail detachment. While lamellipodial and cytoskeletal phenomena are currently the focus of considerable research on cell migration, under many conditions locomotion appears to be rate-limited by events at the cell rear, especially release of cell/s...

Cell migration strategies in 3-D extracellular matrix: differences in morphology, cell matrix interactions, and integrin function.

Cell migration in extracellular matrix is a complex process of adhesion and deadhesion events combined with cellular strategies to overcome the biophysical resistance imposed by three-dimensionally interconnected matrix ligands. Using a 3-D collagen matrix migration model in combination with computer-assisted cell tracking for reconstruction of migration paths and confocal microscopy, we invest...

Distribution and possible interactions of actin-associated proteins and cell adhesion molecules of nerve growth cones.

Actin filaments and their interactions with cell surface molecules have key roles in tissue cell behaviour. Axonal pathfinding during embryogenesis, an especially complex cell behaviour, is based on the migration of nerve growth cones. We have used fluorescence immunocytochemistry to examine the distribution in growth cones, their filopodia and lamellipodia of several actin-associated proteins ...

A talin-dependent LFA-1 focal zone is formed by rapidly migrating T lymphocytes

Cells such as fibroblasts and endothelial cells migrate through the coordinated responses of discrete integrin-containing focal adhesions and complexes. In contrast, little is known about the organization of integrins on the highly motile T lymphocyte. We have investigated the distribution, activity, and cytoskeletal linkage of the integrin lymphocyte function associated antigen-1 (LFA-1) on hu...
